Anastrozole is a non-steroidal aromatase-inhibiting drug approved for treatment of breast cancer after surgery, as well as for metastasis in both pre and post-menopausal women. The severity of breast cancer can be increased by estrogen, as sex hormones cause hyperplasia, and differentiation at estrogen receptor sites. Anastrozole works by inhibiting the synthesis of estrogen.

Anastrozole has been tested for reducing estrogens, including estradiol, in men. Excess estradiol in men can cause benign prostatic hyperplasia, gynecomastia, and symptoms of hypogonadism. It can also contribute to increased risk of stroke, heart attack, chronic inflammation, prostate enlargement and prostate cancer.

Anastrozole binds reversibly to the aromatase enzyme through competitive inhibition, inhibits the conversion of androgens to estrogens in peripheral tissues (extra-gonadal)

Side effects:

The most common side effects of arimidex are hot flashes, nausea, headache, and fatigue. Prescription anastrozole  is only for postmenopausal women. Anastrozole should not be taken if you are pregnant because it may harm your unborn child.
Anastrozole should not be taken with tamoxifen or estrogen -containing therapies.
Based on information from a study of patients with early breast cancer, women with a history of blockages in heart arteries (ischemic heart disease) who take Anastrozole may have a slight increase in this type of heart disease compared to similar patients who take tamoxifen.
Anastrozole can cause bone softening/weakening (osteoporosis)increasing the chance of fractures. In a clinical study in early breast cancer, there were more fractures (including fractures of the spine, hip, and wrist) with Anastrozole (10%) than with tamoxifen (7%).
In a clinical study in early breast cancer, some patients taking Anastrozole had an increase in cholesterol. Skin reactions, allergic reactions, and changes in blood tests of liver function have also been reported.
